The Research of High Dose of Danshen Injection Cause Lung Injury in Mice

Objective: To observe lung tissue injury in mice by high dose of Danshen injection,and discuss the safety of high dose of Danshen injection which provides the reference for the clinical use of Danshen injection. Methods: Thirty mice were randomly divided into control group,low Danshen injection group and high Danshen injection group. The mice of control group were intravenously administered saline. The mice of low and high Danshen injection groups were intravenously administered Danshen injection dose of 0. 64 g / kg and 5. 76 g / kg,respectively. Five days later,lung tissue was observed by HE staining,tumor necrosis factor α( TNF-α) and interleukin-1β( IL-1β) in the peripheral blood serum were measured by ELISA method. Results: There was no abnormal phenomenon in the lung tissue of the mice of control and low Danshen injection groups. The lung tissue of high Danshen injection group showed diffuse hemorrhage and with inflammatory cells infiltration. ELISA results showed that the concentration of TNF-α and IL-1β of high Danshen injection group was significantly higher than control group and low Danshen injection group( P 0. 05). Conclusion: High dose of Danshen injection can cause lung tissue injury and induces inflammatory reaction in mice.
