Effects of rosiglitazone on the expressions of cardiotrophin-1 and nuclear factor-κBP65 in ventricular remodeling after acute myocardial infarction in rats

Abstract

Objective To investigate the effects of rosiglitazone on the expressions of cardiotrophin-1(CT-1) and nuclear factor-κBP65(NF-κBP65) in ventricular remodeling after acute myocardial infarction in rats.Methods Totally 45 male Wistar rats were randomly divided into sham operation group(n=15) and acute myocardial infarction group(n=40).The model of acute myocardial infarction in rats was constructed by ligating the left anterior descending coronary.After 24 hours,30 surviving rats were randomly divided into myocardial infarction control group(n=20) and rosiglitazone group(n=20).Rats in rosiglitazone group were treated with rosiglitazone(gastric lavage,4mg/kg·d) for 8 weeks,while those in myocardial infarction control group and sham operation group received gastric lavage of the same amount of saline for 8 weeks.The expressions of CT-1 and NF-κBP65 were detected by Real-time PCR method and radio immunoassay,respectively.Results After 8 weeks,compared with those in sham operation group and rosiglitazone group,the expressions of CT-1 mRNA and NF-κBP65 in the myocardium of non-infraction zone in myocardial infarction control group were significantly higher(P0.05).The expressions of CT-1 mRNA and NF-κBP65 in the myocardium of non-infraction zone in rosiglitazone group were significantly higher than those in sham operation group(P0.05).Conclusion Rosiglitazone may improve ventricular remodeling after acute myocardial infraction in rats by inhibiting the expression of CT-1 and NF-κBP65.
