Abstract
Objective To investigate the effect of rosiglitazone on the expression of nuclear factor-κBp65(NF-κBp65) and interleukin-1β(IL-1β) in ventricular remodeling after acute myocardial infarction in rats.Methods Forty-five male Wistar rats were randomly divided into sham operation group(group B,n=15) and acute myocardial infarction group(n=30).Rats in acute myocardial infarction group were inflicted by ligating the left anterior descending coronary.After 24 hours,30 survived rats were randomly divided into myocardial infarction control group(group A,n=15) and rosiglitazone group(group C,n=15).Rats in group C were treated with rosiglitazone(gastric gavage,4mg/(kg·d)) for 8 weeks,while rats in group A and B were treated with equal amount of normal saline.The expression of NF-κBp65 and IL-1β were detected by Real Time PCR method and radio immunoassay respectively.Results After 8 weeks,compared with group B and group C,the expression of NF-κBp65 and IL-1β mRNA in the myocardium of non-infraction zone in group A was significantly higher(P0.05).The expression of NF-κBp65 and IL-1βmRNA in the myocardium of non-infraction zone in group C was significantly higher than that in group B(P0.05).Conclusion Rosiglitazone may improve ventricular remodeling after acute myocardial infraction in rats via inhibiting the expression of NF-κBp65 and IL-1β.
