Abstract
Objective To investigate the effect of rosiglitazone on the expression of cardiotrophin-1,angiotensin Ⅱ in ventricular remodeling after acute myocardial infarction in rats.Methods Forty-five male Wistar rats were randomly divided into sham operation group A(n=15) and acute myocardial infarction group(n=30).Rats in acute myocardial infarction group were inflicted by ligating the left anterior descending coronary of rat.After 24 hours,30 survived rats were randomly divided into group B (myocardial infarction control group,n=15) and group C(rosiglitazone group,n=15).Rats in group C were treated with rosiglitazone,gastric gavage,4 mg/(kg·d) for 8 weeks,in group A and group B were treated with drinking water.The expression of cardiotrophin-1 and angiotensin Ⅱwere detected by Real Time PCR method and radio immunoassay respectively.Results After 8 weeks,compared with group A and group C,the expression of CT-1 mRNA and AngⅡ in the myocardium of non-infraction zone in group B was significantly higher(P 0.05).The expression of CT-1 mRNA and AngⅡ in the myocardium of noninfraction zone in group C was significantly higher than that in group A(P0.05).Conclusion Rosiglitazone may improve ventricular remodeling after acute myocardial infraction in rats via inhibiting the expression of CT-1 and AngⅡ.
