Abstract

Objective To observe the function of Ruanganning Decoction and explore its mechanism on prevention and treatment of hepatic fibrosis.Methods CCl4 compound factor was used to induce and set up hepatic fibrosis model in Wistar rats.Normal control group,hepatic fibrosis model group,Ruanganning Decoction prevention group and Ruanganning treatment group were designed.After the end of experiment,liver indexes and liver functions of rats were determined in each group:ALT,AST,index of hepatic fibrosis and hyaluronic acid(HA);the pathological determination of liver tissue and IHC staining of transfer growth factor(TGF-β1)in liver tissue.Results 1.The significant difference in histology presented in the comparison of Ruanganning Decoction prevention group and treatment group with model group respectively in hepatic fibrosis rats(P0.05,P0.01).2.Concerning to liver indexes,the significant difference presented between prevention group and model group(P0.05).The difference was not apparent in treatment group.3.Ruanganning Decoction reduced apparently the levels of ALT and AST in rat serum and HA content in liver tissue of rat,indicating the significant difference compared with model group(P0.01).4.Compared with model group,the treatment in Ruanganning Decoction prevention group and treatment group inhibited obviously the expression of TGF-β1.Conclusion Ruanganning Decoction prevents the occurrence and development of hepatic fibrosis by protecting liver cell,alleviating the injury of liver cell,promoting the degeneration of ECM,reducing the production and release of cell factor TGF-β1.
