Abstract

【Objective】To estimate the cytotoxicity of α-amanitin to 2.2.15 cell lines and then to investigate the inhibitory influence of α-Amanitin on the secretion of HBsAg and HBeAg to 2.2.15 cell lines.【Methods】HepG2 2.2.15 cells were incubated for 3 days with DMEM culture medium containing different concentration of α-Aman-itin and 10% fetal bovine serum,the growth inhibitory effect of α-Amanitin on 2.2.15 cell lines was examined by the MTT assay,the concentration of HBsAg and HBeAg in the culture medium were accessed by TRFIA.【Results】No or little significant cytotoxicity of α-Amanitin to 2.2.15 cell lines was observed in the concentration ranging from 0.0064 μg/mL up to 0.8 μg/mL,while significant cytotoxicity was founded in the concentration ranging from 4 μg/mL up to 20 μg/mL with a TC50 value of 10.19 μg/mL.The concentrations of HBsAg and HBeAg of 2.2.15 cell lines treated by α-amanitin were decreased compared with negative control group,with an IC50 value of 0.495 μg/mL and 0.346 μg/mL,respectively.The inhibitory effection was enhaced with the increased concentration of it.【Conclusions】 At the no or little cytotoxicity concentration,α-Amanitin could suppress the secretions of HBsAg and HBeAg of 2.2.15 cell lines,which suggested that it maybe correlated with inhibiting the duplication of HBV-DNA by α-Amanitin.
