Abstract
In order to screen the ideal method to extract genomic DNA from the leaves of Clematis,9 species of the Clematis were used as the experimental material in this study. Depending upon the index of DNA purity,yield,concentration and time consuming,three methods,CTAB-SDS,simplified CTAB,modified CTAB was compared to achieve the optimal one. The results showed that in the respect of purity,yield,concentration,the order from high to low quality was CTAB-SDS, simplified CTAB and modified CTAB,respectively; and the time consuming order from more to less time was the modified CTAB,CTAB-SDS,and simplified CTAB. The quality of genomic DNA extracted by CTAB-SDS was better than others for PCR amplification template preparing. In conclusion,the CTAB-SDS was the most suitable method for genomic DNA extraction from the leaves of Clematis.
