Abstract
It is the critical first step how to obtain high-quality DNA in molecular biology research.In this paper,their genomic DNA from stem,leaves and endosperm of Pinus armandi Franch were extracted using five methods,which were the classical CTAB method,modified CTAB method,simple CTAB method,SDS micro-extraction method and Zigenhagen method.Then,the extracted DNAs were analyzed by ultraviolet spectrophotometer,agarose gel electrophoresis and standard deviation and standard error of arithmetic mean.The results showed:1The modified CTAB method was appropriate for leaves genomic DNA extraction,whose yield was moderate,changes of standard deviation and standard error were lower,and DNA quality was better and stable;2In stem genomic DNA extraction,the yield and the purity of DNA from the Zigenhagen method were highest and its changes of standard deviation and standard error were lowest.Its DNA quality was obviously better and stable;3For endosperm,the yield and the purity of DNA from simple CTAB method was highest,and its changes of standard deviation and standard error were lowest,which indicated that the DNA quality from simple CTAB method was obviously better and stable.Therefore,different methods should be used to extract genomic DNA in the different tissue of one and the same plant.To obtain the best result,the method of DNA extraction need to be explored in practice.
