Effection of Tonglian Decoction on Cell Proliferation and Cell Cycle of Cell Line MGC803 of Gastric Carcinoma

Abstract

Objective To investigate effect of Tonglian decoction( TD) on proliferation of gastric carcinoma cell line MGC803 through determination on cell morphology and cell cycle; to discuss inhibition mechanism of the prescription on gastric cancer. Methods Human gastric carcinoma cell line MGC803 cultured in vitro treated with TD and its modified formula( THC),and Xiao- ai- ping Injection( XAP) as the positive controlled drug. Cell proliferation is assayed by MTT,cells morphology is observed with microscope and cell cycle is measured with Flow Cytometer( FCM). Results Dose for 50 percent cells inhibition rates( IC 50) of TD is 60. 2 μg·mL- 1. Compared with blank controlled group,TD can inhibit cells morphology and proliferation. In TD group,rate of cells in phase G 2 of cell cycle gets to( 69. 56 ± 2. 06) %. Conclusion TD can be used to regulate MGC803 carcinoma cells proliferation by inhibiting cells entering M phase,having similar mechanism with XAP,which is widely used to anti- gastric cancer in clinic.
