Effects of A-SAA and simvastatin on the expressions of interleukin-6 and -8 of human umbilical vein endothelial cells

Abstract

Objective To observe the effects of acute phase serum amyloid protein A(A-SAA) and simvastatin on the expressions of interleukin (IL)-6 and -8 of human umbilical vein endothelial cells(HUVEC). Methods Total RNA of HUVEC preincubated with A-SAA (1mg/L, 10mg/L) and simvastatin (5μmol/L, 10μmol/L) for 8 hours or 24 hours was isolated by TRIZOL method. Semi-quantitative RT-PCR was performed to quantitate the expressions of IL-6 and IL-8. Results A-SAA induced the IL-6, IL-8 mRNA expressions of HUVEC( P0.01), and the results showed that the A-SAA-induced expressions of IL-6 and IL-8 mRNA were dose-dependent and time-dependent. Simvastatin dereased the A-SAA-induced expressions of IL-6 and IL-8. Conclusion A-SAA increases IL-6 and IL-8 gene expressions, which may contribute to cardiovascular complications. Simvastatin may exert anti-atherosclerotic effect by reducing the expression of IL-6 and IL-8.
