Inhibitory effect of simvastatin on the expression of MCP-1 and IL-6 secreted by HUVECs

Abstract

Objective: To observe the effect of simvastatin on the expression of IL-1,IL-6,MCP-1 and uPA/ uPAR mRNA and MCP-1,IL-6 protein in HUVECs induced with LPS.Methods: Cultivated HUVECs were passaged to the third generation.Then they were divided into four groups randomly: the control group,the LPS(100ng/mLgroup),the simvastatin(5μg/mL) group,the LPS and simvastatin group.The expression of IL-1、IL-6、MCP-1 and uPA/ uPAR mRNA and proteins were measured using real-time quantitative PCR and ELISA.Results: Compared with the control groups,the expression of IL-1、IL-6、MCP-1 and uPA/ uPAR mRNA and proteins in the supernatant were significantly increased in LPS groups(P0.01).Compared with the LPS groups,the expression of IL-1 、IL-6、MCP-1 and uPA/ uPAR mRNA and proteins in the supernatant were statistically decreased in LPS and simvastatin groups(P0.01).Conclusion: Simvastatin can inhibit the synthesis expression of IL-1、IL-6、MCP-1 and uPA/ uPAR in HUVECs,delay the process of atherosclerotic.
