Influence of Jianpi Xiaozheng Formula on Apoptosis and Cell Cycle of Gastric Carcinoma Cell line MGC803

Abstract

Objective:To investigate the effect of Jianpi Xiaozheng formula on proliferation,apoptosis and cell cycle of gastric carcinoma cell line MGC803 in vitro.Methods:After gastric carcinoma cell line MGC803 was treated with Jianpi Xiaozheng Formula in different concentrations,cell proliferation was assessed using MTT assay.The apoptosis rate was examined by flow cytometer using Annexin V-FITC/PI double staining.Apoptosis and cell cycle were examined by flow cytometry(FCM).Results:Jianpi Xiaozheng Formula significantly inhibited the proliferation of MGC803 cells in a concentration-dependent manner.The apoptosis rate of high-concentration group,moderate-concentration group and low-concentration group in 24h were 49.83%,55.29%,16.73% and in 48h were 84.52%,55.58%,51.76%,respectively.As compared with the negative control group,when the cells were treated by high-concentration group,the percentage of the cells in G2-M phase increased in treatment group,which showed the cells were arrested at the G2-M phase of the cell cycle with the increased rate of cell apoptosis.Conclusion:Jianpi Xiaozheng Formula can inhabit the proliferation of MGC803,and can significantly induce apoptosis and arrest the cells at the G2-M phase of the cell cycle.
