Abstract
Objective:To study the effect of Chinese compound Jiedu Xiaozheng Yin on apoptosis related genes of human gastric carcinoma cells.Methods:Cultured human gastric carcinoma cells BGC-823 was divided into four groups: low-dose,medium-dose,high-dose of drug serum group and control group,each group treated with corresponding rat serum containing Jiedu Xiaozheng Yin or control serum.The cells were cultured for 24 hours,48 hours or 72 hours,then Immunofluorescent staining was performed on part of the cells to detect the expression of Bax,Bcl-2,Fas and Fas-L,and results were observed and semiquantitativly analysed via laser scanning confocal microscopy(LSCM).The other cells were prepared and observed via electron microscopy.Results:The serum of rat pretreated with Jiedu Xiaozheng Yin induced apoptosis of gastric carcinoma cells.Compared with the control group,the expression of Bax and Fas was significantly increased(P0.05),while the expression of Fas-L was significantly decreased(P0.05) in the groups with drug serum treatment,and both in a dose dependent manner.The drug serum made no difference on the expression of Bcl-2.Early apoptosis were found via electron microscopy as chromatin margination,mitochondrial crista disappearance and hyperchromation.Conclusion:Jiedu Xiaozheng Yin may regulate the levels of Bax,Fas and Fas-L in gastric carcinoma cells,thus promote cell apoptosis and weaken immune escape.
