Abstract
Objective To study the effects of grape seed procyanidin(GSP) on the expression of nerve growth factor(NGF) after cerebral ischemia reperfusion injury(CIBI) in rats.Methods 48 rats were divided randomly into four groups(n=12):sham operation group,ischemia-reperfusion(I/R) model groups,lose dose GSP(40mg/kg) group,high dose GSP(10mg/kg) group.Reversible middle cerebral artery occlusion(MCAO) model was established by intraluminal suture technique.Sham operation group rats were operated while middle cerebral artery were not blocked.After 2h of occlusion and 48h of reperfusion,using behaviour test,evaluate the neurological deficiency of each group.The expressions of NGF in the cortex were measured by immunohistochemical method.Results The nerves function injuries were severe in model group rats,neurological score of rats treated with GSP was lower apparently compared with model group rats(P0.05);Immunohistochemical studies showed that compared with I/R model group and the expression of NGF was up-regulated remarkedly in GSP group(P0.05).Conclusion The results indicate GSP could improve the expression of NGF after cerebral ischemia reperfusion.It may be one of mechanisms for the treatment of ischemic stroke.
