Abstract

Objective To observe the protective effect and mechanism of effective components group of Xiaoshuantongluo(XECG)on ischemia/reperfusion(I/R)injury in rats. Methods Healthy male adult Sprague Dawley rats were randomly divided into the sham operation group, the model group, the nimodipine(4mg·kg-1)group,and high, middle and low dose XECG groups(200,100,50mg·kg-1).Middle cerebral artery occlusion was adopted to establish the rat ischemia/reperfusion model. After ischemia for 120 min and reperfusion for 24 h,the pathological injury of the ischemia side was observed by HE staining.Expressions of tumor necrosis factorα(TNF-α),Interleukin 1β(IL-1β)were detected by western blotting technique. Results XECG could significantly improve the pathological changes of cortical penumbra brain tissues,Compared with the I/R model group,XECG could significantly inhibit expressions of TNF-α、IL-1β.Conclusion XECG has obviously protective effect on Sprague Dawley rats after cerebral ischemia and reperfusion, its mechanisms may be associated with inhibiting inflammatory, expression of TNF-αand IL-1βproteins.
