Optimization of Influencing Factors on Adventitious Bud Formation in Pepper Regeneration

Abstract

The effects of different explants,combinations of hormones and concentrations of AgNO_3 on adventitious bud differentiation were investigated by using the variety of pepper(Capsicum annuum L)No.1 Hajiaoas experimental material,and the cotyledon,hypocotyl and cotyledon with petiole as explants. The optimal mediums for adventitious bud induction of the three explants were obtained.The orthogonal test result showed that on the adventitious bud differentiation period,the combination of 6- BA and IAA could obviously raise the differentiation rate,the ratio of BA/IAA had a direct effect on the adventitious bud differentiation,and 6-BA 6 mg/L plus IAA 2 mg/L could give the highest induction rate.The AgNO_3 significantly increased the adventitious bud induction rate and shortened the time for the bud formation.In addition,the bud induction rate of cotyledon with petiole was much higher,next came to cotyledon and then hypocotyl among the three explants.The optimal medium compositions for adventitious bud were MS + 6-BA 6 mg/L+ IAA 2 mg/L+ AgNO_3 4 mg/L and pH5.8,and the adventitious bud differentiation rate reached 88.2%.
