Induction of the Growth Inhibition and Apoptosis in Human Lung Cancer A549 Cell Line by 5-ally-7-gen-difluormethylenochrysin

Abstract

Objective To investigate the effect of 5-ally-7-gen-difluormethyleno-chrysin(ADFMChR) on the growth and apoptosis of human lung cancer A549 cells cultured in vitro.Methods A549 cells were treated with various concentrations of ADFMChR.Cell proliferation of A549 cells was evaluated by colony formation in plate and in soft agar test.The apoptosis of A549 cells induced by ADFMChR was determined by DNA fragment assay and flow cytometry with PI staining.Results Data of colony formation in plate and in soft agar test indicated that ADFMChR could inhibit the growth of A549 cells in a concentration-dependent manner.DNA fragment assay indicated that DNA ladder bands could be shown in DNA agarose gel electrophoresis after treatment with ADFMChR at 10μg/mL for 48h.PI staining flow cytometry showed that the apoptosis of A549 cells was induced by ADFMChR in a concentration-dependent manner.Conclusions ADFMChR posseses induction of the growth inhibition and apoptosis of A549 cells in vitro.
