Abstract

Objective:To investigate the effect of 5-Allyl-7-gen-difluoromethylenechrydsin(ADFMChR) on growth inhibiting in cervical cancer cell cultured in vitro. Methods: Human cervical cancer(HeLa) cells and human endodermis of allantoic veins(HUVEC) cells were cultured in vitro. The trypan blue exclusion method was used to investigate the growth inhibitory effect of ADFMChR in HeLa cells and,.HUVEC cells;Clone-formation assay in plate and soft agar was used to test inhibition of clone formation of HeLa by ADFMChR.Flow cytometry(FCM) using PI staining was used to analyse the cell cycle change after treatment with ADFMChR.Results: ADFMChR inhibited proliferation and growth of HeLa cells in a dose and time-dependent manner, its efficacy was stronger than that of Chrysin. While there was no significant effect in HUVEC cells,ADFMChR could inhibit the growth of HeLa cells in a concentration-dependent manner.FCM analysis indicated that the differdrent density ADFMChR effect on HeLa cells, can significantly arrest the cell cycle in G1 phase. Conclusion: ADFMChR posseses the effective of restrainting the growth of human cervical Cancer Cell Cultured in vitro.
