Expression of vascucar endothelial growth factor in gliomas

Abstract

Objective To study the expression of vascular endothelial growth factor(VEGF) mRNA in gliomas, and the neovascularization of tumor tissues, and the correlation between them. Methods The mRNA level of VEGF in surgical samples from 49 glioma patients was examined by using semi quantitative reverse transcription polymerase chain reaction(RT PCR). Three glioma cell lines and 12 samples of normal brain tissues obtained from cranicerebral injury surgery were also studied. Immunohistochemical SP method was used to analyze the microvessel density(MVD) in tissues. Results The expression level of VEGF mRNA was significant high in gliomas, the positive rate was 65.3%. The expression level paralleled with the histopathologie grade of glioma. HUVEC and 3 glioma cell lines also expressed high level of VEGF, but the normal brain tissues remained lower expression. The MVD increased with glioma pathologic grades. There was a positive correlation between mRNA level of VEGF and MVD. Conclusion VEGF mRNA,as well as MVD had an important significance to evaluate the malignant biological behavior of glioma cells. The overexpression of VEGF resulted in vascular proliferation of gliomas, and played a critical role in the invasion of tumor tissues.
