Abstract
Objective:To establish an RP-HPLC method for determination of chlorogenic acid and bacalin in Yinhuang granules.Method: The analysis was performed on a Kromasil C18 column(4.6 mm×250 mm,5 μm) with acetonitrile and water containing 0.4% phosphoric acid as the mobile phase in gradient elution. The flow rate was set at 1.0 mL·min-1.The UV detector wavelength was at 325 nm,and the column temperature was at 30 ℃.Result: The linear ranges of chlorogenic acid and bacalin were 0.060 0-1.20 μg(r = 0.999 9) and 0.272-5.44 μg(r=0.999 9);the average recoveries of chlorogenic acid and bacalin were 99.5% and 97.9%,and RSD were 0.73% and 1.79% respectively. Conclusion: The method was simple, feasible, and it can be used for the quality control of Yinhuang granule.
