Abstract
Objective To explore the effect of trimetazidine on myocardial fibrosis in diabetic cardiomyopathy rats.Methods Forty-five Wistar rats were randomized into two groups:normal control group(n=8)and experimental group(n=37).The rats in experimental group were fed with high sugar-fat diet for 8 weeks to induce the insulin resistance,then were injected with STZ for 4 weeks.After that,the rats were fed for 16 weeks,and then the model rats of diabetic cardiomyopathy with FPG≥16.7 mmol/L were divided into two groups:DM group(n=18) and trimetazidine group(n=15).The rats in trimetazidine group received trimetazidine for 4 weeks,while the rats received same volume of distilled water instead in the other two groups.Then all rats were killed.The interventricular septa were formalin-fixed,paraffin-imbedded and HE-stained.The microstructure of cardiac muscle was observed under transmission electron microscope(TEM).Expression of collagen-Ⅰ and Ⅲ in the cardiac muscle was detected.All data were analyzed statistically.Results The positive expression of collagen-Ⅰand Ⅲ in DM group was significantly higher than in trimetazidine group.The microstructure of cardiac muscle in DM group was in great disorder,and cardiac muscle fibers were ruptured obviously.In trimetazidine group,the cardiac muscles were damaged slightly,and the microstructures were in normal order.Conclusion Trimetazidine can improve the pathological damages of cardiac muscle by influencing the myocardial energy metabolism of diabetic cardiomyopathy.
