Abstract

Objective To investigate the effects of trimetazidine,compared with perindopril,on myocardial energy metabolism and ultrastructure in chronic heart failure(CHF) rats.Methods Seventy-two male SD rats were randomly divided into control group and model group.Rats in model group were injected subcutaneously with isoproterenol.Four weeks later,the rats in model group were randomly divided into three groups:untreated group(group M),trimetazidine treated group(group T) and perindopril treated group(group P).After treatment for five weeks,echocardiography and myocardial pathological examination were performed.The energy metabolism indexes of myocardium were determined.Results Compared with the rats in group M,EF and FS of the rats in group T increased by 6.5% and 11.8% respectively.The level of ADP,AMP,ATP/ADP and TAN(total adenosine) had no significant differences among groups T,P and control.Group T showed a significant increase in myocardial sarcoplasmic reticulum Ca2+-ATPase activity as compared with group M,but there were no significant differences among groups T,P and control.The microscopic and electron microscopic examination showed that the myocardial injury in groups T and P was ameliorated as compared with that in group M.Conclusion Trimetazidine can improve myocardial energy metabolism,pathological change and ultrastructure and enhance the cardiac function in HF rats.
