Abstract
Objective To investigate the protective effects of rosiglitazone on rat kidney fibroblasts(NRK) damaged by high glucose.Methods The NRK cells were cultured in vitro,and were divided into five groups: normal glucose group(NG,1000 mg/L D-glucose),high glucose group(HG,4500 mg/L D-glucose),HG+RGZ(5μmol/L),HG+RGZ(10μmol/L) and HG+RGZ(15μmol/L).The mRNA expressions of TGF-β1 and PAI-1 were measured using RT-PCR.Results Compared with NG group,mRNA expressions of TGF-β1 and PAI-1 increased significantly in HG group,while rosiglitazone inhibited those changes in dose-dependent manner.Conclusion Rosiglitazone can ameliorate the damage on the kidney fibroblasts induced by high glucose,thus have protecting effects on kidney.
