Abstract
Aim: To investigate the protective effects of rosiglitazone on rat kidney fibroblasts (NRK) damaged by high glucose.Methods:The NRK cells were cultured in vitro,and then divided into five groups: normal glucose group(NG, 1 000 mg/L D-glucose), high glucose group(HG, 4 500 mg/L D-glucose), HG+RGZ(5 μmol/L), HG+RGZ(10 μmol/L) and HG+RGZ(15 μmol/L). The mRNA expressions of TGF-β_1 and PAI-1 were measured using RT-PCR.Results: Compared with NG group, mRNA expressions of TGF-β_1 and PAI-1 increased significantly in HG group, while rosiglitazone inhibited those changes in a dose-dependent manner.Conclusion: Rosiglitazone can ameliorate the damage on the kidney fibroblasts induced by high glucose, thus have protecting effects on kidney.
