Effection on CYP2D1 Expression in Liver and Brain by Dimethylaminoazobenzene

Abstract

Objective To investigate the effection on cytochrome P450 2D1(CYP2D1) mRNA expression in liver cancer and brain tissues of rats induced by dimethylamineoazobenzene(DAB).Methods The expression of CYP2D1 mRNA in these tissues was detected by promoted RT PCR.Results After 30 cycles was carried out in the first PCR,the expression of CYP2D1 mRNA was detectable in liver tissues of control and hepatocarcinogenesis rats,but not detectable in brain tissues.After another 30 cycles in the second PCR,with the product of the first PCR as templat,then both in liver and brain tissues the expression of CYP2D1 mRNA could be detectable,but the intensity of electrophoresis band of PCR products of brain tissues were weaker compared to liver tissues.Conclusion CYP2D1 mRNA expression level in hepatocarcinogenesis rat liver tissues is same to in normal rat ,CYP2D1 mRNA also can express in brain tissues and its level is lower in liver tissues.So dimethylamineoazobenzene could not induce the expression of CYP2D1 mRNA in liver and brain.
