Abstract

Objective:To evaluate the effect of a novel anti-human DR5 monoclonal antibody(mAb mDRA-6) on the apoptosis of human hepatocellular carcinoma cell lines. Methods: mDRA-6 cytotoxicity on cell lines was detected by MTT analysis. The influence of mAb mDRA-6on the morpha of cells lines was observed under fluorencence microscope. The rate of apoptosis was detected by flow cytometry with Annexin V-FITC/PI staining. Results: Cells treated with mDRA-6 exhibited typical apoptosis features in morphology.Viability of cells decreased with mAb concentration increasing by MTT analysis. The apoptosis rate of the SMMC-7721 cells was 35% in the presence 2 mg/L mDRA-6 for 6 hours by flow cytometry analysis with Annexin V-FITC/PI staining. Conclusion: mDRA-6 could induce SMMC-7721 cells apoptosis and has apoptosis activity.
