Abstract

The operating condition of aqueous two-phase systems(ATPS) was optimized to extract xylanase from Bacillus pumilus,using response surface analysis.The optimal system contained NaCl 6 %,PEG2000 18 % and K2HPO4 10 %,respectively.Resulting in a portion factor of xylanase and protein 3.38,0.13,respectively,a purification factor of 7.93 and a 84.24 % yield of enzyme activity.The characteristics of the partially purified xylanase were also studied and results showed that the optimal temperature and pH of it was 50 ℃ and 7.0,respectively.
