Abstract
In order to explore a fast,simple,and especially reliable cotton genomic DNA extraction method,two traditional simple methods were improved and compared in experiment.The results showed that the improved SDS-CTAB method was characteristic of clean solution and DNA high yield,but there was more experiment steps in the method.The improved CTAB method has fewer steps,and was suit to large number of samples extraction of multiple batch cycle process,but DNA yield was lower and solution was not clean.The results demonstrated that two improved method,comparing to traditional CTAB methods of total DNA isolation in cotton,can get quickly sufficient pure genomic DNA,which was sufficient for PCR screening in transgenic descendant detection.
