Abstract

CTAB method and SDS method two methods of cotton genomic DNA extraction have been introduced in this paper. Different treatments on cotton leaves have also been compared according to the effect of DNA extraction. The results show that high-quality DNA can be obtained by both methods. In addition, the fresh cotton leaves and the leaves stored under 4℃ and room temperature for 1 day are all suitable for DNA extraction. If the cotton leaves have been exposed in the 42℃ environment for more than one day, the DNA in them is apparently degraded. However, it is no problem for satisfying the PCR analysis.
