Abstract

Objective: To establish a simple and sensitive high-performance liquid chromatography coupled with evaporative light-scattering detector ( HPLC-ELSD) method for the determination of four saccharides including fructose,glucose,mannotriose and stachyose in Danshen Injection.Methods: The HPLC method was conducted on a Prevail Carbohydrate ES column( 250 mm × 4.6 mm,5 μm) with acetonitrile-water( 67 ∶33) as mobile phase.The flow rate was 1.0 mL·min-1 ,the drift tube temperature of ELSD was set at 35 ℃,and carrier flow was 3.0 L·min-1.The column temperature was 35℃.Results: The calibration of fructose,glucose,mannotriose and stachyose were linear over the ranges of 375.6~187 8,233.0~116 5,228.4~114 2 and 300.2~150 1 μg·mL-1 respectively, and the average recoveries were 102.4% ( RSD,2.18% ) ,103.9% ( RSD,3.45% ) ,103.1% ( RSD,2.56% ) , 100.9% ( RSD,2.71% ) respectively.Conclusion: The method is accurate,simple and reproducible for quality control of Danshen Injection.
