Abstract

Objective:To study the effect of 5-fluorouracil and oxaliplatin on Fas Ligand mRNA expression of human colon cancer cells line SW480 and apoptosis of lymphocyte induced by it with cell culture. Methods:Different concentrations of 5-fluorouracil and oxaliplatin have been used in the cultured human colon cancer cells line SW480.The change of Fas Ligand mRNA was detected by the reverse transcription polymerase chain reaction method(RT-PCR)and apoptosis rate of lymphocyte induced by SW480 cell was measured by flow cytometry analysis. Results:Results of RT-PCR revealed that the higher concentration 5-fluorouracil or oxaliplatin raised Fas Ligand mRNA expression on SW480 cell; Results of flow cytometry showed that the higher concentration 5-fluorouracil or oxaliplatin increased apoptosis rate of lymphocyte induced by SW480 cell, the comparison of different concentrations groups of 5-fluorouracil or oxaliplatin had statistical significance. Conclusion:5-fluorouracil ane oxaliplatin all can upregulate functional Fas Ligand mRNA expression of human colon cancer cells line SW480 and the upregulative effect can assist cancer cells to counterattack lymphocyte.
