Abstract
Objective:To observe the effect of ligustrazine on protein expression of bone marrow cell of blood deficiency mice induced by radiation and to study the molecular mechanism of ligustrazine in blood enriching function.Methods:The blood deficiency mice were induced by using a single dose of 3.5 Gy ~(60)Co γ radiation.Myeloid colony cell was cultured and the number of CFU-GM,BFU-E,CFUE and CFU-mix were observed and counted.By using proteomics methods,such as two-dimensional electrophoresis(2-DE),image analysis,in-gel digestion,and mass spectrometry with bio-informatics,the differential expression proteins were separated,analyzed and identified.Results: After irradiation,colony number of CFU-GM,BFU-E,CFU-E and CFU-mix decreased significantly.Ligustrazine can raise significantly the colony number of CFU-GM,BFU-E,CFU-E and CFU-mix of mice bone marrow and reverse partly the change of protein expression induced by irradiation,which makes five kinds of down-regulated proteins raise again and five kinds of up-regulated proteins fall back.Conclusion: Ligustrazine can promote hemopoietic progenitor cell proliferation of bone marrow of blood deficiency mice induced by radiation and regulate the expression of many kinds of bone marrow cell proteins.The latter effect could be one of the main mechanisms that ligustrazine can promote hemopoietic cell growth and proliferation.
