【Objectives】 To detect the effects of bFGF on proliferation of human periodontal ligament cells in vitro,thus to explore the mechanisms of bFGF effecting orthodontic tooth movement.【Methods】 Human periodontal ligament cells were cultured in vitro,and the 5th passage of cells planted in 50 wells of a 96-well culture plate and every well contained 4×103 cells.They were divided into 10 groups and every group contained 5 wells.According to bFGF concentration,no bFGF group,0.5 ng/mL bFGF group,1.0 ng/mL bFGF group,2.0 ng/mL bFGF group,4.0 ng/mL bFGF group 8.0 ng/mL bFGF group,16.0 ng/mL bFGF group,32.0 ng/mL bFGF group,64.0 ng/mL bFGF group and 128.0 ng/mL bFGF group were named respectively.When all groups were being in exponential phase of growth under microscope,they were dyed by MTT method.Then OD value of every well was calculated by enzyme linked immunodetection meter.【Results】 By contrast to OD mean of control group,OD mean of every group increased gradually from 0.5 ng/mL to 16.0 ng/mL of bFGF concentration(P 0.05,0.01),and reached the peak level on 16.0 ng/mL of bFGF concentration.From 32.0 ng/mL to 128.0 ng/mL of bFGF concentration,OD means of every group decreased step by step(P 0.05,0.01).【Conclusion】 bFGF can play an important role in orthodontic rebuilding of the periodontal membranes.bFGF with lower concentration can promote the growth of human periodontal ligament cells,however higher concentration can inhibit the growth of human periodontal ligament cells.
