Effect of Basic Fibroblast Growth Factor on Proliferation and Dose-effect Relationship of Tendon Cells in Vitro

Abstract

Objective To study the effects of different concentration basic fibroblast growth factors(BFGFs)on proliferation and dose-effect relationship of tendon cells cultured in vitro,and to estimate the optimal BFGF concentration of promoting proliferation.Methods The groups of different BFGF concentrations were stained by MTT method and their average OD values were detected by enzyme linked immunodetection meter.Results Compared with the OD mean value of the control group,the BFGF groups of 1.0,2.0,5.0,10,30,40,and 50 ng/ml showed significant difference(P0.05),while the BFGF groups of 0.5 and 2.0 ng/ml showed no significant difference(P0.05).When BFGF concentration varied between 0~5.0 ng/ml,the OD mean value increased gradually;while BFGF concentration exceeded 5.0~50 ng/ml,it decreased step by step.Conclusions 1.0,2.0,5.0,and 10 ng/ml BFGF could promote proliferation of tendon cells,0.5 and 20 ng/ml BFGF show no significant effect on proliferation of tendon cells,and 30,40 and 50 ng/ml BFGF could inhibit proliferation of tendon cells.The results suggest that 5.0 ng/ml is the optimal BFGF concentration of promoting proliferation.
