HPLC quantitative analysis of three flavonoid glycosides in Folium Apocyni Veneti and Folium Poacyni

Abstract

Objective: To establish an HPLC method for the determination of three flavonoid glycosides in Folium Apocyni Veneti and Folium Poacyni.Methods: The analysis was performed on an Elite-ODS(5 μm,150 mm×4.6 mm) column with the mobile phase consisted of acetonitrile-water-tetrahydrofuran-acetic acid(8∶ 81∶ 10∶ 1) at a flow rate of 1.0 mL·min-1,the detection wavelength was 258 nm and the column temperature was 30 ℃.Results: The linear ranges of rutin,isoquercitrin and quercitroside were 0.026-0.520,0.094-1.880 and 0.084-1.680 μg with the correlation coefficients of 0.9996,0.9997 and 0.9990,respectively.The average recoveries(n=5) were 97.5%,98.4%,97.7% with RSDs of 1.9%,1.8%,2.2%.Difference in the distributions and contents of isoquercitrin from two herb medicines was also shown from HPLC analysis.Conclusion: This method is simple,sensitive and rapid,which can be used for the quality control of Folium Apocyni Veneti and Folium Poacyni.
