Abstract

To study the extraction and Separation of chlorogenic acid from leaves of Eucommia ulmoides. Effects 0f Water and different concentiration of ethanol, methanol, acetone on the yield of chlorogenic acid were discussed. Main factors influencing the extraction of chlorogenic acid were studied by orthogonal experiments and resins were used to separate chlorogenic acid .The results appeared: Water was better than the other three solvents to extract chlorogenic acid at the temperature of 50℃, and the yield was 1.06%, so water was used as solvent to extract chlorogenic acid.Optimized conditions for the extraction of chlorogenic acid were obtained, extraction temperature 60℃, ratio of material to liquor 1:16, pH 4, extraction time 3h. The selected GC-I resin was optimum absorbent to separate chlorogenic acid, the pH is maintained at 3, flow rate of adsorption is 3BV/h. The experiment shows that the purity of resulted product was arrived to 30.88% and the yield was 76.51%.
