Abstract
Objective:To investigate the reversal effect of isorhynchophylline on multidrug resistance of human adenocarcinoma of lung cell line A549/DDP.Methods:The effects of isorhynchophylline on the proliferation of A549 and A549/DDP cells were observed by MTT assay.The inhibitory effects of cisplatin(DDP),vincristine(VCR),adriamycin(ADM) and etoposide(VP-16) used alone or in combination with isorhynchophylline on the proliferation of A549 and A549/DDP cells were evaluated by MTT assay.VCR induced apoptosis were determined by flow cytometry(FCM).The effects of isorhynchophylline on DDP accumulation in A549/DDP cells was determined by ICP-MS.Results:Isorhynchophylline at the concentration of 1.5mg·L-1 and below showed no significant cytotoxicity to A549 and A549/DDP cells.The resistances of A549/DDP cells to VCR,VP-16,ADM and DDP were 19.58,11.97,21.74 and 24.29 times as those of A549 cells.After 0.5mg·L-1 isorhynchophylline was added,the chemo-sensitivity of A549/DDP cells to VCR,VP-16 and DDP was increased as 1.27,1.81 and 2.19 times as that of A549 respectively.After 1.0 and 1.5mg·L-1 isorhynchophylline was added,the chemo-sensitivity of A549/DDP cells to VCR,VP-16 and DDP was increased significantly.1.5mg·L-1 isorhynchophylline enhanced significantly the VCR-induced apoptosis.0.5,1 and 1.5mg·L-1 isorhynchophylline increased DDP accumulation in A549/DDP cells by 1.79,2.46 and 9.30 folds as those of control group.Conclusion:Isorhynchophylline could moderately reverse the multidrug resistance of A549/DDP cells through increasing the accumulation of chemo-drugs in cells and promoting apoptosis.
