Abstract

ObjectiveTo investigate the protective effect of silybin-phosphatidylcholine compound(SPC) on kidney in STZ-induced diabetic rats.MethodsFifty SD rats were fed for one week,then ten rats were randomly chosed as control group,the other forty rats were injected with 60 mg·kg-1 streptozotocin(STZ) through enterocoelia respectively.The levels of blood-fasting sugar of the rats were measured after 72 hours,the thirty-three rats whose blood glucose were higher than 16.7 mmol·L-1 were randomly divided into three groups:model group,high dose SPC group and minidose SPC group(n=11).The rats in normal control and model groups were administered with 1% sodium carboxymethyl cellulose(CMC-Na) suspension for 12 weeks,and two SPC groups were respectively administered with 300 mg·kg-1 and 150 mg·kg-1 SPC mixed with 1% CMC-Na suspension for 12 weeks.At the end of 12 week,the levels of serumal creatinine(Cr),blood urea nitrogen(BUN) and fructosamine(FMN) were detected,at the same time,the levels of MDA,SOD and advanced glycation endoproducts(AGEs) in the nephridial tissue of cortex renis were detected.ResultsCompared with control group,the weights of diabetic rats were decreased and the kidney indexes were increased in model group(P0.001).The weights of diabetic rats in different doses of SPC groups were obviously increased and the kidney indexes were decreased than those in model group(P0.05).Compared with control group,the serum levels of Cr,BUN,FMN and MDA in nephridial tissue were obviously increased,SOD activity were obviously decreased in model group(P0.001),and in SPC groups,these indexes were obviously altered than those in model group(P0.05).Conclusion SPC possesses potential protective effect on kidney in rats with diabetes.
