Inhibitory Effect of Baicalim on Human Melanoma A375 Cell Line

Abstract

Objective:To investigate the inhibitory effect of Baicalim on human Melanoma A375 cell line in vitro.Methods:Melanoma A375 cells were cultured invitro and then treated with different concentrations(0.1~50μmol/L) of Baicalim solution.Cells were observed with inverted microscope,the growing curve was drawn.MTT assay was adopted to determine the inhibition rate of Baicalim on melanoma A375 cells;Flow cytometry was used to detect the apoptosis of melanoma A375 cells.Results:The growing curves and MTT assay showed that different concentrations of Baicalim were cytotoxic to melanoma A375 cells in a time-dependent and dose-dependent manner.The FCM analysis showed that the apoptosis rate of melanoma A375 cells was increased after treatment of Baicalim.Meianoma A375 cells presented the characteristic morphological changes of apoptosis under inverted microscope.Conclusion:Baicalim could inhibit the proliferation of human melanoma A375 cells invitro,which way associated with cell apoptosis.
