Effect of bFGF on expression of HSP70 mRNA in cerebral tissue during cerebral ischemia in rats

Abstract

Objective To investigate the expression of HSP70 mRNA in hippocampus and cortex after cerebral ischemia and reperfusion in rats,and to explore regulative effect and mechanism of basic fibroblast growth factor(bFGF) on HSP70 mRNA in brain tissue.Methods The model of middle cerebral artery occlusion was performed with intraluminal filament blockade.The expression of HSP70 mRNA and cell apoptosis in hippocampus and cortex was detected with RT-PCR method and TUNEL method.Results The expression of HSPT0 mRNA in hippocampus and cortex was increased after cerebral ischemia and reperfusion.The expression of HSP70 mRNA in bFGF group was increased,but cell apoptosis was decreased.Conclusion The results indicate that bFGF depress cell apoptosis and participate in the regulation of expression of HSP70 mRNA in ischemic neurons.
