Abstract

Objective To evaluate the effects of aminoguanidine(AG)in prolonged hemorrhagic shock. Methods 28 rabbits subjected to prolonged hemorrhagic shock and resuscitation were randomly divided into hemorrhagic shock group (14 rabbits)and AG group(14 rabbits,AG was administered as soon as resuscitation was performed). The plasma levels of endothelin(ET),tumor necrosis factor α(TNF α),interleukin 6(IL 6),Iinterleukin 8(IL 8)and nitric oxide(NO)were detected at preshock, postshock, just after resuscitation and 0 5h,2h,4h after resuscitation. The survival rates of 24h and 48h were recorded. Results The plasma levels of ET,TNFα,IL 6,IL 8 and NO were markedly higher after shock than those of before shock, and maintained at high level.Compared with shock group, lower plasma levels of ET,TNFα,IL 6,IL 8 and NO,as well as higher survival rate were observed in AG group. Conclusion The ET,TNFα,IL 6,IL 8 and NO play an important role in the development of hemorrhagic shock.AG used as a selective inhibitor of the inducible isoform of nitric oxide synthase can help to improve the outcome of prolonged hemorrhagic shock.
