Abstract

Objective To observe the effects of glutathione on the abnormal behavior,nigral antioxidation system,mitochondrial respiratory-chain function and neuron morphology in model rat of PD induced by 6-hydroxydopamine (6-OHDA). Methods The rats of model PD were established through stereotoxic microinjection of 6-OHDA. The 24 rats of model after behavior test were randomly divided into 3 groups:Model group(MG),Glutathione group (GG) and sham-operated group (SG). Every group was given relevant treatment for 45 days. All rats behavior were tested before and after giving medicine . The activity of GSH-Px,content of MDA,ROS and Mitochondrionase complex I of nigra were assayed after giving medicine.The nigra form was observed by immunohistochemistry. Results (1)GSH had no effect on its rotation behavior. (2)Glutathione could decrease the nigral oxidative stress of PD rats model. (3)Glutathione could exert some protective effect on mitocondrial respiratory-chain. (4)The TH-IR neurons of GG increased in injected side compared with MG. Conclusion GSH could decrease the nigral oxidative stress of PD model rats,and exert some protective effect on mitocondrial respirtory-chain and neuron morphology.
