Abstract
Objective To explore the effect and mechanism of nerve growth factor(NGF) and calcitonin gene related peptide(CGRP) on the expression of C/EBP homologous protein(CHOP) protein in parietal lobe cortex of rats with focal cerebral ischemia/reperfusion(I/R).Methods The middle cerebral artery occlusion(MCAO) was employed to make focal cerebral ischemia model.The expression of CHOP was investigated by immuohistochemical and Westtern blotting methods and analyzed by metamoph analysis system.Results The expression of CHOP protein in parietal lobe cortex was absent in sham group,but found 12h,reached a maximum 24h,markedly decreased 72h after reperfusion in I/R group,and the expression of CHOP protein was markedly higher in I/R group compared with sham group(P0.01).The expression of CHOP protein was lower in NGF group or in CGRP group than in I/R group respectively(P0.01),and markly lower in NGF and CGRP group than in I/R group(P0.01),and lower in NGF and CGRP group than in NGF group or CGRP group respectively(P0.01).Conclusion NGF and CGRP downregulate the expression of CHOP protein and have coordinate effects on the parietal lobe neurons following focal ischemia/reperfusion.
