Abstract
Taking the stem-segment with axillary buds as explants,the effects of mediums with different concentration of plant growth regulator on inducing cluster buds,multiplication coefficient and rooting rate were studied,the effect of different medium without agar on rooting in vitro were also studied.The results showed that the optimal medium for inducing cluster buds was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L+sucrose 30 g/L,pH 5.8,and the ratio of induction was 92.17%,The rooting culture medium was 1/2MS+IBA 0.1 mg/L and the ratio of rooting was 93.23%.In all kinds of mixture media without agar pearlite-vermiculite-peat was better,and it was more suitable for transplanting compare to agar media.
