Abstract

Objective To investigate the effect of Saikosaponin D on apoptosis in human cervical cancer Hela cell line and its detailed mechnisms.Methods Human cervical cancer Hela cells was treated by Saikosaponin D with various concentrations(0、2.5、5、7.5μg/ml respectively),then the cell viability was determined by MTT assay at different time.After treatment for 48h,apoptosis rate was determined by flow cytometry.Cell total proteins were also extracted after treatment by Saikosaponin D for 48h,and western blot was performed to detect the expression of survivin.Results Saikosaponin D inhibited human cervical cancer Hela cell line in a time-and concentration-dependent manner.After treatment by Saikosaponin D for 48h,apoptosis rate was increased paralleled with the concentration of Saikosaponin D.Western blot indicated that Saikosaponin D suppressed the expression of survivin.Conclusions It is suggested that Saikosaponin D induces apoptosis of human cervical cancer Hela cell line,and this effect may be partly mediated by decreases in survivin protein expression.
