Determination of chlorgenic acid and caffeic acid in powder injection of Dandehon herb by HPLC

Abstract

OBJECTIVE To establish an HPLC method for the determination of chlorgenic acid(CHA),caffeic acid(CA) in powder injection of Dandehon herb.METHODS DikmaTM C18 column(200 mm × 4.6 mm,5 μm) was adopted.The mobile phase was a mixture of acetonitrile-0.1% phosphoric acid solution(17:83) and the flow rate was 1.0 mL·min-1.The detection wavelength was 326 nm and column temperature was controlled at 30 ℃.RESULTS For chlorgenic acid,the method had good linear relationship within the range of 0.108-3.464 μg·mL-1 with the correlation coefficient 0.9998 and the averge recovery(n = 9) was 99.68%.For caffeic acid,the method had good linear relationship within the range of 0.114-3.648 μg·mL-1 with the correlation coefficient 0.9999 and the averge recovery(n = 9) was 98.73%.CONCLUSION The established HPLC method is sensitive,simple,accurate and reproducible.It can be used for the determination of chlorgenic acid,caffeic acid in powder injections of Dandehon herb.
