Abstract
Using Dendrobium adventitious buds as materials, according to single-factor randomized block design and orthogonal experiment method, under the conditions of 25℃, 2 000 lx, 12 h·d-1, the effect of basic medium, NAA, 6-BA, BR and sucrose on the proliferation of adventitious buds were examined. The results showed that: 1/2 MS was the best base medium for Dendrobium adventitious buds proliferation; NAA was a major factor in the proliferation of bud multiplication and rooting. The combination(1/2 MS + 3 g·L-1No.1 Precious + NAA 1.0 mg·L-1+6- BA 1.5 mg·L-1+ BR 0.001 mg·L-1+ sucrose 30 g·L-1+ envelope protein hydrolyzate 0.25 g·L-1) was conducive to proliferate of adventitious buds. The bud proliferation rate( 17.31 ± 0.09) and rooting shoots multiplication rate(5.68 ± 0.03) reached at the 1% significant level than others.
