Abstract
BT type rice cytoplasmic male sterility line were compared with its maintainer,restorer and F 1 hybrid using mitochondrial DNA (mtDNA) RAPD analysis.The results showed that difference was between not only the cms line and its maintainer,but also cms line and its F 1 hybrid.Primer OPJ-08 generated a polymorphic product of 800 bp in sterile line,and primer OPK-10 amplified a specific fragment of 900 bp in F 1 hybrid.The two fragments were then cloned into plasmid vector and converted to RFLP probes.Southern hybridization patterns revealed polymorphism between sterile line and F 1 hybrid using OPJ-08 800 fragment as probe,and between sterile line and maintainer line using OPK-10 900 fragment as probe.We infered that the two mtDNA fragments may be associated with cms in rice.
