Study on plant regeneration from cotyledon explants of a melon cultivar 'Huangdanzi'

Abstract

To establish plant regeneration technique system of a melon cultivar 'Huang danzi',and to realize its genetic ability and character in regeneration,cotyledon pieces from yang seedling of 'Huangdanzi' were inoculated on bud inducing medium MS+6-BA 2.0 mg/L.It proved that the inducing bud(s) from cotyledon explants were adventitious bud(s) source.58 pieces of cotyledon explants regenerated adventitious buds at an average frequency of 44.8%,and produced calli in different characters and amounts.On medium MS+6-BA 0.05 mg/L,the adventitious buds elongated,but part of them exhibited vitrifica-tion.In the medium 1/2 MS+IAA 0.5 mg/L,the elongated shoots rooted at the rate of 99%.Compared with other thick-skin melon cultivars in relative reports,'Huangdanzi' does not regenerate easily,but obvious differences existed between individuals in regenerative rate and characters.Probably we can select individuals with higher regenerative ability from 'Huangdanzi',and breed them to new lines with better regeneration ability.
