Abstract

The cotyledons and hypocotyls of Tianyuan No.6tomato were used as explants to study the effects of the kind of different hormones and their combinations on callus induction,callus subculture,differentiation of adventitious buds and roots and the impact of factors on the tomato callus browning.The results showed that medium of MS+IBA 0.8 mg/L+6-BA 2.0mg/L could induce the best callus,the medium of MS+IBA 1.0mg/L+6-BA 0.5mg/L was most suitable for callus subculture,the mediums of MS+IAA 0.5mg/L+6-BA 2.0mg/L and MS+IAA 0.5mg/L+6-BA 1.0mg/L were the best mediums respectively for inducing buds from explants directly and callus,and the medium of MS+IAA 0.5mg/L was the ideal medium for the induced rooting from adventitious buds.Then kind of explants and illumination would affect the browning of the callus in some way.But the 10mg/L VC and 0.3activated carbon could slow it.
